The construction of this volume has been guided by two personal convictions. Experience in the field of experimental chemotherapy, both in the pharmaceutical industry and academia, has convinced us that recent quantum technological advances in biochemistry, molecular biology, and immunology will permit and, indeed, necessitate an increasingly greater use of rational drug development in the future than has been the custom up to now. In Part l, therefore, we asked our contributors to provide detailed reviews covering the biology of the malaria parasites and their relation with their hosts, the experimental procedures including culture techniques that are necessary to take a drug from primary screening to clinical trial, and an account of antimalarial drug resistance. Our second conviction is that many research workers are all too loath to learn from the lessons of the past. For this reason we asked the contributors to Part 2 of this volume to review very thoroughly the widely scattered but voluminous literature on those few chemical groups that have provided the antimalarial drugs in clinical use at the present time. Much can be learned from the history of their development and the problems that have arisen with them in man. This study is concerned with the preparation of: a) peroxides based on qinghaosu, b) peptide pro-drug derivatives of primaquine, and c) thymidylate synthetase inhibitors as potentially new antimalarial agents. During this study, thirty (30) target compounds were prepared and submitted for antimalarial evaluation. The thirty compounds included twenty-four (24) qinghaosu analogs, three (3) primaquine derivative and three (3) thymidylate synthetase inhibitors. Twenty-one of the qinghaosu analogs and all three of the thymidylate synthetase inhibitors were evaluated in the malaria in vitro drug screen. All but three of the qinghaosu analogs showed activity in this screen, whereas the thymidylate synthetase inhibitors were inactive. The three peptide pro-drug derivatives of primaquine were tested for radical curative activity against Plasmodium cynomologi in rhesus monkeys, All three compounds showed cures greater than that expected for the primaquine content of each pro-drug. Twenty-three target compounds were evaluated for blood schizonticidal suppressive activity against P. berghei in mice, and ten compounds were tested for causal prophylactic activity against P. berghei yoelii in mice. Interest in the 4-amino-1,5-naphthyridines as potential prophylactic antimalarial agents has been generated by the structural similarity of the ring system to both the 4- and 8-aminoquinolines. In synthetic studies during the program, the authors sought to expand upon the derivatives of 4-amino-1,5-naphthyridine which are currently available in an effort to secure an optimal prophylactic antimalarial drug. Accordingly, as a result of the synthetic efforts, a total of 26 naphthyridine target and 145 naphthyridine intermediates and miscellaneous precursors have been prepared and submitted for biological testing. While antimalarial testing has not yet been completed, results obtained for most of the target drugs submitted to WRAIR have disclosed no significant prophylactic activity in the lower dosage range.
