Chitosan, a natural based polymer obtained by alkaline deacetylation of chitin, is non-toxic, biocompatible, and biodegradable. These properties make chitosan a good candidate for the development of conventional and novel drug delivery systems. Recently, there has been a growing interest in the chemical modification of chitosan in order to improve its solubility and widen its applications. Chemical modification of chitosan is useful for the association of bioactive molecules to the polymer and controlling the drug release profile. Chemical modification will introduce desired properties and enlarge the field of the potential applications of chitosan with the choice of various types of side chains. In this monograph, recent studies on the various types of chitosan microspheres are discussed from the viewpoint of drug delivery applications. Moreover, different types of chitosan derivatives developed as controlled drug delivery carriers and their preparation methods are discussed in detail. The modifications discussed here include amphiphilic chitosan derivatives, N-(aminoalkyl) chitosan, carboxymethyl chitosan, thiolated chitosans, phosphorylated chitosan, etc. Special emphasis has been given to the development of chitosan-based polyelectrolyte complexes, graft copolymerized chitosan and chitosan-based stimuli-responsive hydrogels as drug delivery carriers. The reviewed studies lead to the conclusion that chitosan derivatives are promising materials for the controlled delivery of various types of therapeutic agents.

Conventional dosage forms often lead to wide swings in serum drug concentrations. Most of the drug content is released soon after administration, causing drug level in the body to rise rapidly, attain maximum concentration and then decline sharply. This sharp fluctuations often cause unacceptable side effects at the peaks, followed by inadequate therapy at the troughs. In the transdermal drug delivery system, drug is released at a constant rate and provides plasma concentration that remains constant over an extended period of time. In the present work, chitosan film has been used as rate controlling membrane for the transdermal drug delivery of an antiemetic drug, Ondansetron HCl. This book throws light on the preparation and evaluation of chitosan membrane as well as provides a new method for the preparation of chitosan gel.This book also, provides the way of fabrication and evaluation of transdermal patches utilising chitosan films which shows promising potential for use in the transdermal delivery of Ondansetron HCl.

Chitin is the second most abundant polysaccharide present in nature; however, chitin has more applications when transformed into chitosan (CS). It is biocompatible, biodegradable, mucoadhesive, soluble in acidic-solutions, nontoxic and nonallergenic. The main drawback of chitosan in pharmaceutical procedures is its low solubility in physiological medium. Chitosan shows physicochemical characteristics that allow it to interact with a wide variety of molecules. This is of particular interest when increasing the solubility of poor water-soluble drugs. For this purpose, chitosan can be used in oral, nasal and ocular routes. In order to modulate drug release rate and achieve a proper drug delivery in physiological medium, some parameters can be modified when solid dispersions or nanoparticles (NPs) based on chitosan are being designed. In case of nanoparticles, chitosan can be used as the main component or as a modifying agent. In order to optimize drug loading and drug delivery, response surface methodology (RSM) is an interesting tool usually underestimated in the pharmaceutical field, which allows us to optimize the parameters involved in the process simultaneously and not by different steps, which usually lead to mistakes.
