Morphometric studies reveal that the volumes of many brain structures differ between clinical and non-clinical populations. Specifically, populations characterized by affective disturbances related to heightened emotional reactivity and diminished emotion regulation have been found to possess smaller volumes of brain structures involved in emotion reactivity and regulation. I sought to determine whether similar associations would be found between brain structure and trait emotion reactivity and regulation in healthy individuals. Specifically, I predicted that emotion reactivity, as measured by negative affect, would be negatively related to the volume of the ventral anterior cingulate cortex, insula, orbitofrontal cortex, and amygdala, and that emotion regulation, as measured by usage of expressive suppression and cognitive reappraisal, would be positively related to the volume of the dorsal and ventral anterior cingulate cortex, dorsolateral prefrontal cortex, hippocampus, orbitofrontal cortex, cerebellar vermis, and anterior insula. I tested these predictions using two complementary methods, region of interest (ROI) and voxel-based morphometry (VBM). In Study 1, which used a sample of healthy adult women, trait usage of expressive suppression was correlated positively with the volume of the middle frontal gyrus and anterior insula, and negatively with ventral anterior cingulate volume. Trait usage of cognitive reappraisal was correlated positively with the volume of the bilateral dorsal anterior cingulate cortex. All of these correlations were found using both ROI and VBM methods. In Study 2, which used a more diverse sample of healthy males and females ranging in age from 10 to 22, self-reported levels of trait negative affect was correlated negatively with posterior insula volume, although this finding was not confirmed using VBM. Both ROI and VBM revealed that trait suppression usage frequency was correlated positively with right ventral anterior cingulate and anterior insula volume. The strength of the relationship between suppression and anterior insula volume increased as subject age increased. Although no ROIs were found to correlate with trait reappraisal usage frequency, VBM found that reappraisal positively correlated with gray matter clusters in the prefrontal cortex. The findings from this dissertation demonstrate that the structural alterations seen in individuals suffering from mood and anxiety disorders may not result from psychopathology per se, but may instead be on the lower end of the continuum of emotion reactivity, emotion regulation, and associated brain structure.

A characteristic of the cingulo-thalamic or the subicular thalamic pathway had been investigated during the discriminative avoidance conditioning in which a CR of stepping in an activity wheel prevents a footshock 5 sec. after an auditory CS+. Enhanced neuronal discharges in the mediodorsal (MDN) and the anterior ventral (AVN) thalamic nuclei were found in rabbits with the combined lesions of posterior cingulate cortex and subiculum in Experiment 1 as after the separate lesions in a previous study (Gabriel et al, 1987). The frequency of CR was also greater in rabbits with the combined lesions during early training. This result suggests that both cingular and subicular input limits the firing of thalamic neurons. Experiment 2 tested the hypothesis that hippocampal lesions will mimic the effects of subicular lesions. However, the hippocampal lesions enhanced the unit activity in MDN and posterior cingulate cortex (Area 29c/d and Area 29b) but not in AVN or anterior cingulate cortex. The hippocampal lesions did not increase CR-frequency during acquisition, extinction or during reacquisition. These results suggested a suppressive modulation by hippocampal efferents on several limbic structures and a functional independence of subiculum and hippocampus. In Experiment 3, neuronal activity in the hippocampus, AVN, and posterior cingulate cortex in the well trained rabbits were recorded after temporary hippocampal disruption by medial septal injections of the local anesthetic(Tetracaine:TH) or GAB$sb{rm A}$ agonist (muscimol: MUS). The CS evoked discharges in CA3, CA1, and DG were attenuated and the theta-like, sinusoidal macropotentials were altered following either TH or MUS injection. Spontaneous unit activity of CA3 and AVN were also reduced as was the incidence of CR. These findings suggest that disturbing the function of the hippocampus has devastating effects on the performance of the active avoidance task in rabbits in comparison to permanent lesions in the hippocampus. These findings indicate that the hippocampal modulation is limiting or suppressive on the limbic thalamus and the limbic cortex and that the interaction between the hippocampus and other limbic structures is involved in the performance of the active avoidance task.

Research implicates the Anterior Cingulate Cortex (ACC) mediates the affective component of pain processing but not spatial learning. The Posterior Cingulate Cortex (PCC) mediates spatial learning but not pain processes. This study investigated the roles of each cortex in spatial learning and pain processing. Lesions were made using standard stereotaxic procedures on 96 animals. Inflammatory condition was induced via carageenan. Animals were randomly assigned between both MPWT and PEAP followed by Morris Swim Test or vice versa in order to prevent an order effect. As hypothesized, ACC lesioned animals resulted in no spatial learning decrement, but showed no avoidance behavior in PEAP within the inflammatory condition. PCC lesioned animals resulted in no spatial learning decrement and showed no significant avoidance behavior in PEAP that goes against the hypotheses.
