With the recent approval of the first eosinophil-depleting therapeutic agents targeting the IL-5 pathway for treatment of severe eosinophilic asthma, eosinophils and eosinophilic disorders are in the limelight. Indeed, setbacks during clinical development of these compounds have revealed how much remains to be known about eosinophil biology in vivo, and have nurtured profuse research both on basic eosinophil biology and on pathogenic disease mechanisms, in order to better delineate the most meaningful targets for innovative therapeutic strategies. On one hand, variable degrees of eosinophil depletion observed in some compartments during IL-5-targeted treatment indicate that certain eosinophil subsets may not rely on this cytokine and/or that other important pro-eosinophilic mediators and signaling pathways are operative in vivo. On the other hand, it is increasingly clear that disorders involving eosinophils such as asthma are the final outcome of complex interactions between diverse cell types and mediators, beyond eosinophils and IL-5. These include type 2 helper T (Th2) cells and innate lymphoid cells, mast cells, and a variety of factors that either activate eosinophils or are released by them. Although a considerable amount of research has focused on asthma because it is a common condition and because management of severe asthma remains a major challenge, several rare eosinophilic disorders with more homogenous features have proven to be extremely useful models to reach a better understanding of the involvement of eosinophils in tissue damage and dysfunction, and of the micro-environmental interactions operating within the complex network of eosinophilic inflammation. Unraveling this interplay has resulted in advances in the development of molecular tools to detect disease subsets and to monitor therapeutic responses, and in identification of promising new therapeutic targets. This Research Topic dedicated to eosinophilic conditions covers aspects of the biology of eosinophils and closely related cells of particular relevance for drug development, reports on translational research investigating pathogenic mechanisms of specific eosinophilic disorders in humans that will likely result in significant changes in the way patients are managed, and presents an overview of the current advancement of targeted drug development for these conditions, with a special focus on asthma.

Blood Cell Biochemistry was initially conceived as part of the Plenum series Subcellular Biochemistry, from which it has developed into a separate series. This monograph on the "Ultrastructure of Human Sella Tumors" is in fact a study of the correlations of clinical findings and morphology. It is a timely and eagerly awaited publication because of the increasing interest of the endocrinologist in pituitary disorders and of the neuro surgeon in the newest aspects of surgery on pituitary tumors, and also because of the unsatisfactory but still widely accepted classification into eosinophil, basophil and chromophobe pituitary adenomas. This old classification has been mainly based on granule color as seen after hematoxylin-eosin staining, but it does not, after extensive clinical observations, reflect in many instances the type of clinical picture observed. The author has brilliantly succeeded in demonstrating in a convinc ing way, by an extensive study of the relevant literature and by his own histological work, that further insight into the biology of the hypophysis and of the pituitary tumor can be obtained only by newer methods of light microscopic staining, immuno-histochemistry and electron microscopy combined with radio-immunological blood hormone determination.

Thus far in the history of biology, two, and only two, fundamental principles have come to light that pervade and unify the entire science-the cell theory and the concept of evolution. While it is true that recently opened fields of inves tigation have given rise to several generalizations of wide impact, such as the universality of DNA and the energetic dynamics of ecology, closer inspection reveals them to be part and parcel of either of the first two mentioned. Because in the final analysis energy can act upon an organism solely at the cellular level, its effects may be perceived basically to represent one facet of cell me tabolism. Similarly, because the DNA theory centers upon the means by which cells build proteins and reproduce themselves, it too proves to be only one more, even though an exciting, aspect of the cell theory. In fact, if the matter is given closer scrutiny, evolution itself can be viewed as being a fundamental portion of the cell concept, for its effects arise only as a consequence of changes in the cell's genetic apparatus accumulating over geological time. Or, if one wishes, the diametrically opposite standpoint may be taken. For, if current concepts of the origin of life hold any validity, the evolution of precellular organisms from the primordial biochemicals must have proceeded over many eons of time prior to the advent of even the most primitive cell.
