Potassium ion (K+) channels are ubiquitous components and are widely distributed on the surface of the cell membrane in various tissues. There are many types of potassium ion channels (voltage-gated K+ channels, ATP-sensitive K+ channels, Ca2+-activated K+ channels, etc.), and each type of potassium ion channel has its own characteristic features concerning electrophysiology and molecular biology in each potassium ion channel. The potassium ion current across the plasma membrane is considered to have two main physiological functions, one for determining the membrane potential and the other regulating the electrical activity. Membrane hyperpolarization would reduce the excitability of the membrane by shifting the membrane potential away from the threshold for excitation and would inhibit, where it existed, calcium ion (Ca2+) influx through voltage-gated Ca2+ channels by closing the Ca2+ channel. Outward potassium ion currents activated upon depolarization would counteract the depolarizing action of inward currents carried either by sodium ion (Na+) or Ca2+. Therefore, potassium ion channels show the physiological effects in each tissue via membrane potential mediated Ca2+ dynamics (for example, excitation-contraction coupling in muscle). Potassium ion channel activity is also essentially regulated by the intracellular signal transduction pathways, such as receptors (G proteins) second messenger molecules processes in the physiological condition. On the other hand, the modification of potassium ion channel activity would be expected to have a significant effect on the excitability of various cells. Therefore, augmentation of outward potassium ion currents may be useful to treat various diseases (asthma, chronic pulmonary obstructive disease, hypertension, diabetes mellitus, glaucoma, arrhythmias, heart failure, epilepsy, etc.). Furthermore, potassium ion channels may be a target protein for the research and development of a therapeutic agent for various diseases concerning the lungs, heart, nerves, eyes, and pancreas. However, there are few books described in detail about potassium ion channels. This book will focus on the characteristics, function and regulation of several types of potassium ion channels, focusing on the role of these channels not only in terms of physical responses, but also in the aforementioned diseases.

Membrane Receptors, Channels and Transporters in Pulmonary Circulation is a proceeding of the 2008 Grover Conference (Lost Valley Ranch and Conference Center, Sedalia, Colorado; September 3-7, 2008), which provided a forum for experts in the fields of those receptors, channels and transporters that have been identified as playing key roles in the physiology and pathophysiology of the pulmonary circulation. The book rigorously addresses: i) recent advances in our knowledge of receptors, channels and transporters and their role in regulation of pulmonary vascular function; ii) how modulation of expression and function of receptors, channels and transporters and their interrelationships contribute to the pathogenesis of pulmonary vascular disease; and iii) the therapeutic opportunities that may be revealed by enhancing our understanding of this area. The overall goal was to explore the mechanisms by which specific receptors, channels and transporters contribute to pulmonary vascular function in both health and disease, and how this knowledge may lead to novel interventions in lung dysplasia, pulmonary edema, lung injury, and pulmonary and systemic hypertension to reduce and prevent death from lung disease. Membrane Receptors, Channels and Transporters in Pulmonary Circulation is divided into six parts. Part 1 (Ion Channels in the Pulmonary Vasculature: Basics and New Findings) is designated for basic knowledge and recent findings in the research field of ion channels in pulmonary circulation. There are five chapters in Part I discussing the function, expression, distribution and regulation of various ion channels present in pulmonary vascular smooth muscle cells and how these channels are integrated to regulate intracellular Ca2+ and cell functions. Part II (TRP Channels in the Pulmonary Vasculature: Basics and New Findings) is composed of five chapters that are exclusively designed to discuss the role of a recently identified family of cation channels, transient receptor potential (TRP) channels, in the regulation of pulmonary vascular tone and arterial structure. Part III (Pathogenic Role of Ion Channels in Pulmonary Vascular Disease) includes four chapters that discuss how abnormal function and expression of various ion channels contribute to changes in cell functions and the development of pulmonary hypertension. Part IV (Receptors and Signaling Cascades in Pulmonary Arterial Hypertension) consists of five chapters devoted to the role of bone morphogenetic protein receptors, Notch receptors, serotonin receptors, Rho kinase and vascular endothelial growth factor receptors in the development of pulmonary arterial hypertension. Part V (Receptors and Transporters: Role in Cell Function and Hypoxic Pulmonary Vasoconstriction) includes four chapters designed to illustrate the potential mechanisms involved in oxygen sensing and hypoxia-induced pulmonary vasoconstriction and hypertension. Maxi calcium-activated potassium channels (BK) are an amazing category of ion channels which are found in cellular plasma membranes as well as in membranes of intracellular organelles. The function of these channels is to repolarize any excited membrane by passing a potassium outward current, in response to depolarization and/or increase in local calcium levels. Thus, voltage and calcium ions are involved in gating the channel under physiological conditions. This dual activation makes them perfect sensors for many cellular events that require integration between intracellular calcium levels and electrical signals. A plethora of physiological and pathophysiological functions, such as membrane hyperpolarization, modulation of synaptic transmission, hormone secretion or mental deficiencies, vaso-regulation, epilepsies, heart diseases, myotonic dystrophies, hypertension etc, in almost all cells and tissues were reported for these channels. BK channels are main targets for important ligands like alcohol and gaseous neurotransmitters, such as NO, CO or H2S, to name a few. In the last years, the molecular entities and mechanisms involved in modulation of BK channels have gained tremendous attention, as the key role of these channels in cellular processes became increasingly recognized. Indeed, accessory proteins such as slob, beta and gamma subunits, all serve to modulate the channel gating characteristics. Moreover, channel subunit expression and function is further tuned by phosphorylation/ dephosphorylation processes, redox mechanisms and the lipid microenvironment of the BK channel protein complex. This e-book contains structural and functional aspects of BK channels, channel modulation by a variety of agents and cellular components, as well as the channel’s relevance in health and disease.
