Obesity is one of the largest medical epidemics our society faces today. There are over forty obesity-associated diseases that affect nearly every organ system in the body, including Type 2 Diabetes (T2D) and Non-Alcoholic Fatty Liver Disease (NAFLD). Insulin resistance is one of the hallmark characteristics underlying metabolic diseases, and mitochondrial dysfunction is believed to be a central player in this phenomenon, yet our understanding of the specific molecular mechanisms that serve as cause and consequence of defective mitochondria remain complex and elusive despite significant attention. In the studies presented in this dissertation, liver tissue isolated from a rat model of obesity-induced NAFLD and human subject-derived skeletal muscle cells revealed two distinct mechanisms by which mitochondrial dynamics may play a key role in regulating the progression of insulin resistance and cellular bioenergetics in obesity-related metabolic diseases. First, we found that markers of mitochondrial fusion and biogenesis are upregulated in the liver of obese Sprague-Dawley rats following gastric bypass surgery, and expression of a specific fusion marker, namely mitofusin 1, is associated with reductions in body weight and improved insulin sensitivity. Utilizing human primary skeletal muscle cells isolated from percutaneous needle biopsy, we found that metformin treatment in vitro upregulates glycolytic pathways, suppresses mitochondrial respiration, and activates a specific protein of mitochondrial fission. Together, the work presented here describes mitochondrial mechanisms related to insulin resistance in two obesity models and major metabolic tissues.

Both organelles interact at contact sites, defined as mitochondria-associated membranes (MAMs), and disruption of ER-mitochondria interactions was recently involved in hepatic (1) and muscle (2) insulin resistance and alterations of glucose homeostasis (3) and that organelle miscommunication was associated with insulin resistance in various mice models of obesity and T2D (1-4) . However, the role of MAMs in u00df cells is still largely unknown and their implication in glucotoxicity-associated u00df cell dysfunction remains to be defined. Beta cell dysfunction in type 2 diabetes (T2D) is associated with glucotoxicity-mediated alterations of both mitochondria and endoplasmic reticulum (ER). However, the role of MAMs in u00df cells is still largely unknown and their implication in glucotoxicity-associated u00df cell dysfunction remains to be defined. Here, we report that acute glucose stimulation increases ER-mitochondria interactions and Ca2+exchange in INS-1E cells, whereas disruption of MAMs alters glucose-stimulated insulin secretion (GSIS). Conversely, chronic exposure (48h-72h) of of either human pancreatic islets or INS-1E cells to moderate glucotoxicity (16.5 and 22.5mM glucose, respectively) altered GSIS. This is associated with a reduction ER Ca2+ content, an increase of basal mitochondrial Ca2+ content and reduced ATP- stimulated ER-mitochondria Ca2+ exchanges, despite an increase of ER-mitochondria interactions, measured by both electronic microscopy and proximity ligation assay. Furthermore, glucotoxicity-induced perturbations of Ca2+ signalling are associated with ER stress, altered mitochondrial respiration and mitochondria fragmentation, and these organelle stresses may participate to increased organelle tethering, as a protective mechanism. Lastly, sustained induction of ER-mitochondria interactions using a linker reduced organelle Ca2+ exchange, induced mitochondrial fission and altered GSIS. Therefore, dynamic organelle coupling participates to GSIS in u03b2 cells and over-time disruption of organelle Ca2+ exchange might be a novel mechanism contributing to glucotoxicity-induced u00df cell dysfunction. Therefore, we propose that MAM integrity could be dynamically and differentially regulated in u00df cells during the progression of T2D pathology, highlighting that MAM regulation plays an important role in the control of u00df cell function. 1.
