Virus-host interactions are complex and have been a driving force of the everlasting evolutionary progress of both virus and host. Ever since the first virus infection took place, it has been an arms race of defense and counter defense between virus and host. To further uncover the underpinnings of this arms race, plant RNA viruses were utilized to uncover the mechanisms of virus-host interactions. Three projects were chosen that focus on different aspects of the viral interaction. Specifically, three separate projects investigated viral-host relationships in suppression activities of the viral proteins, coat protein's trans effect on recombination, and co-encapsidation of host-RNA. The first project focused on the host effect on the virus through the RNA silencing system that can degrade viral RNA. Additionally, the possibility that virus-host interactions might provide an evolutionary advantage in favor of the host was investigated.

The research seeks to understand the virus-host plant interactions for cassava brown streak disease (CBSD) caused by two viruses, Cassava brown streak virus (CBSV) and Ugandan Cassava brown streak virus (UCBSV) of the genus Ipomovirus, family Potyviridae. The diversity of six CBSD isolates from the endemic (Kenya, Malawi, Mozambique and Tanzania) and the recently developed epidemic areas (Uganda) of the disease in eastern Africa was studied. Five cassava varieties differing in virus resistance levels; Albert, Columbian, Ebwanateraka, TMS60444 (all susceptible) and Kiroba (tolerant) were graft-inoculated with the UCBSV and CBSV isolates. Based on a number of parameters, the isolates can be grouped into two main categories; severe and milder forms. Transmission of viruses using non-vector modes confirmed that CBSV was sap transmissible from cassava to cassava. Graft-inoculation of infected scions onto CBSD-free cassava plants was the most efficient mode of transmission which resulted in 80 and 100% rate for UCBSV and CBSV respectively. The two virus isolates were not transmitted through contaminated tools and hands. The effect of host-tolerance on virus was investigated in a long-term experiment where three cassava varieties Albert, Kiroba and Kaleso (field-resistant to CBSD) were graft-inoculated with UCBSV and CBSV. The three cassava varieties showed differences in virus movement, symptom development, severity and relative virus titres. The mechanisms of resistance to CBSD were investigated by making cuttings, from various parts of the plants, and a greater number of disease-free plants were generated from cuttings made from Kaleso than Kiroba and Albert. The fecundity of B. tabaci and its ability to transmit the virus were determined and results indicated no significant differences in the ability of the three cassava varieties to support whitefly development. Finally, thermal and chemical treatments of tissue cultured plants were conducted and the combinations of both treatments produced the greatest number of disease-free plants in all three varieties; Kaleso (50%), Kiroba (44%) and Albert (35%). The information generated in this thesis has greatly improved our understanding of the interactions between the three biotic factors; the host, virus and vector in the CBSD-pathosystem, which would be highly useful in designing effective disease management strategies.

The domestication of grapes dates back five thousand years ago and has spread to nearly all continents. In recent years, grape acreage has increased dramatically in new regions, including the United States of America, Chile, Asia (China and India), and Turkey. A major limiting factor to the sustained production of premium grapes and wines is infections by viruses. The advent of powerful molecular and metagenomics technologies, such as molecular cloning and next generation sequencing, allowed the discovery of new viruses from grapes. To date, grapevine is susceptible to 64 viruses that belong to highly diverse taxonomic groups. The most damaging diseases include: (1) infectious degeneration; (2) leafroll disease complex; and (3) rugose wood complex. Recently, two new disease syndromes have been recognized: Syrah decline and red blotch. Losses due to fanleaf degeneration are estimated at $1 billion annually in France alone. Other diseases including leafroll, rugose wood, Syrah de cline and red blotch can result in total crop loss several years post-infection. This situation is further exacerbated by mixed infections with multiple viruses and other biotic as well as adverse abiotic environmental conditions, such as drought and winter damage, causing even greater destruction. The book builds upon the last handbook (written over twenty years ago) on the part of diagnostics and extensively expands its scope by inclusion of molecular biology aspects of select viruses that are widespread and economically most important. This includes most current information on the biology, transmission, genome replication, transcription, subcellular localization, as well as virus-host interactions. It also touches on several novel areas of scientific inquiry. It also contains suggested directions for future research in the field of grapevine virology.
