Retroviruses arguably belong to the most fascinating of all viruses because of their unusual and highly efficient mode of replication involving reverse transcription and integration of the viral genome and a complex system of transcriptional and post transcriptional regulatory mechanisms. The retroviral membrane shows selective enrichment in sphingomyelin and cholesterol, the hallmark components of lipid rafts. The mechanism by which the virus obtains this unique lipid composition is unclear. Using thin layer chromatography analysis of the RSV lipid envelope, I demonstrate that in the absence of Env, Gag virus-like-particles (VLPs) retain a raft-like lipid composition. Furthermore, the envelopes of VLPs produced from a dually acylated Gag protein are even more raft-like. However, I also demonstrated that RSV particles are not resistant to cold Triton X-100. These results suggest that multiple types of protein-lipid interactions determine the viral lipid composition and that a raft-like lipid composition does not necessarily correspond with detergent resistance.
